Buoyant Titration of Bovine Mercaptalbumin Chemically Modified at the Carboxyl Groups

چکیده

Bovine mercaptalbumin has been modified at the carboxyl groups by means of I-ethyl-3 (3-dimethylisopropyl) carbodiimide and glycine amide. A stable derivative was obtained after modification of 54 groups. The modified protein differed only slightly from the unmodified one with respect to the optical rotatory dispersion parameters ao, bo and ~. This preparation was studied in the analytical ultracentrifuge in a CsCI gradient in the pH-interval from 2 to 1 I. The buoyant densities were determined and compared with those of the unmodified protein in the same pH-interval. The contribution from the carboxyl groups to the buoyant density is discussed and compared with results obtained with other proteins and synthetic polypeptides.

Moving boundary electrophoresis behavior and acid isomerization of human mercaptalbumin.

The behavior of bovine plasma albumin in moving boundary electrophoresis has been studied in great detail, especially by Aoki and Foster (l-6) and by Cann (7-10). In general terms, it has been found that this protein yields a single moving boundary at pH values above the isoelectric point (approximately pH 4.5) but two, and in some cases three, boundaries between pH 3.5 and 4.5. These results h...

Quantitative Determination of Carboxyl Groups in Cellulose by Complexometric Titration

Complexometric titration was used to determine the carboxyl group content of a series of oxidatively treated (selective and non-selective oxidation) nature cellulose fibers (cotton). The results were correlated with those obtained from the conventional methylene blue method; both methods show an excellent correlation. The progress of the cellulose fibers oxidation was monitored using viscosimet...

Titration behavior of histidines in human, horse, and bovine hemoglobins.

The titration curves of the human hemoglobins AZ and F, the slow and fast components of horse hemoglobin, and bovine hemoglobin B have been analyzed, mainly to obtain information about the titration behavior of the histidines in these hemoglobins. The results were also compared with earlier data concerning human hemoglobin A and bovine hemoglobin A. In both hemoglobin As and F, 18 titratable hi...

The heterogeneity of bovine albumin with respect to sulfhydryl and dimer content.

A relatively fresh bovine plasma albumin (BPA) preparation (0.65 sulfhydryl group per albumin molecule; 5 to 6% dimer) and an old bovine mercaptalbumin (BMA) preparation (0.72 sulfhydryl group per albumin molecule; 8 to 9 % dimer) were fractionated in a phosphate gradient on DEAE-Sephadex A-SO. Two dimer fractions were obtained. One dimer fraction had a relatively high sulfhydryl content and co...

Bovine Mercaptalbumin and Non-mercaptalbumin Monomers

The monomer macroheterogeneity of commercial bovine serum albumin preparations is entirely due to oxidation of the mercaptalbumin sulfhydryl group. When the sulfhydryl group is oxidized to oxidation states higher than disulfide, circular dichroism and fluorescence data show that a tryptophanyl and a cystinyl residue are perturbed. Measurements conducted through the pH range of the N-F transitio...
